The third installment in the Assassin's Creed franchise, this game's story picks up right after the events of Assassin's Creed II, showing Ezio Auditore traveling to Rome to recruit a new force of assassins. Brotherhood is the first game in the series to offer online multiplayer.

I don't have this game so I'm probably wrong, but I thought the Templars in multiplayer were all disguised as Assassins as part of an Animus training program.
Ezio is contrasted with the others on the cover because they are his enemies. He has his wrist blades drawn and raised, giving him an unfriendly pose as everyone else regards him from behind his back, which is also unfriendly. Even more notably, Ezio is shown in full color, while the others are in black and white, again contrasting the two as opposites (protagonist vs antagonists).
Because the PR people who were in charge of the game's box art don't actually know the specifics of the story and were just given a bunch of assets and told to do market research to see what tested well.
...that Ezio totally stabs these motherfuckers in the Templar missions in the singleplayer campaign? The very first guy you murder is the Butcher/Executioner dude.
Could always ask the guy who designed the boxart. He's a member on this site, and he's posted the other box covers he's done as well. I can't recall his name atm, though.
