Originally a carpenter named Jumpman, this Italian plumber has gone on to become the most recognizable video game character of them all, starring in a veritable pantheon of titles like kart racing and sports. He has been voiced by Charles Martinet for nearly 20 years.
Luigi is Mario's brother. He began as a simple palette swap of Mario, but was later differentiated. In his later forays, he provides comic relief.
Princess Peach Toadstool is usually the damsel in distress from the Mario franchise. She gets kidnapped frequently by Bowser, though she's not averse to being captured by various other villains from time to time.
Bowser is one of the most iconic villains in gaming. His role as the main antagonist in the Mario franchise has cemented this King of Koopas' status in video game history.

Donkey Kong Jr. is, or at least has been considered, the son of the original Donkey Kong, who made his debut in the 1983 arcade classic which bore his name. He is actually a younger version of the current Donkey Kong.
Disk-kun (or Diskun) was the mascot of the Famicom Disk System.
The Original Donkey Kong made his debut in the game of the same name, where he tossed barrels at his nemesis Mario. His grandson, Donkey Kong Jr., went on to create a legacy of his own in games such as Donkey Kong Country and Donkey Kong Country Tropical Freeze.
Mario's original girlfriend back in Donkey Kong. We're guessing it didn't work out.
One of the main characters in the Famicom Tantei Club franchise, Ayumi is a member of the games' titular club. Ayumi is also a trophy in Super Smash Bros. Melee.

Medusa is often portrayed in games as a creature that is a cross between a woman and a snake. The lower body is often a snake tail, and the hair is usually a mass of snakes. Other than this, the head and torso are that of a woman.
Zelda is the titular figure of The Legend of Zelda. She is most often depicted as the princess of Hyrule.
Lakitu has appeared in a variety of Mario games; from Super Mario Bros on the NES right up to Mario Kart 7. He is responsible for dropping Spinies onto the levels below, holding up signs and his very own start signal from all nine Mario Kart games.
Link is the iconic protagonist of The Legend of Zelda franchise.
The right-hand man of Count Dracula, Death is easily recognized by his giant scythe and ominous cloak. Not the same Death who appears in other games.
The embodiment of evil in the Zelda series, Ganon lusts after the Triforce and the ultimate power it affords its user. He often appears in his less monstrous form, Ganondorf.
Twinbee is the ship from the popular Twinbee series of shoot-em ups. It has also made a cameo in the Parodius series as well.
Taizo Hori is the protagonist of the Dig Dug series, as well as a recurring character in the Mr. Driller series. Though he solved the Dig Dug incident at a young age, he worries that he has been overshadowed by his son Susumu.
Impa is the caretaker of Princess Zelda and a member of the Sheikah race. Like Link and Zelda, Impa has appeared in numerous incarnations throughout the Zelda timeline.
Winbee is an anthropomorphic ship that appears in the Twinbee series of games.
Donbe is one of two main characters of the Shin Onigashima series.
An enemy from the Dig Dug series and a supporting character from the Pac-Man World series.
A member of the titular Tantei Club in the Famicom Tantei Club franchise, as well as the main protagonist of the first two games.
