I also found the segments during of the end of Deadly Premonition to be pretty sad, aswell. =(
" Finding David in the Mass Effect 2 Overlord DLC "Not sure that was my saddest moment. They foreshadowed it a little too much, which took away from the final reveal. But I did think it was one of the stronger moments in ME2, and made up somewhat for the almost total lack of character interaction otherwise. I like the regret on the brothers face at what he had done.
I think the ending of MGS: Peace Walker was pretty sad. It fills in the last important gap in the story of Naked Snake, and locks him into his tragic destiny as the "villain" of the future games.

I have to agree, I loved ME2 but I didn't think the David reveal was very shocking. It was far too obvious and the DLC itself wasn't long enough to build up any tension or feelings for the characters.
I'd probably go with the ending of Red Dead Redemption. Even if you knew it was going to happen, the actual events were pretty brutal and very graphic. I can't imagine anyone wasn't at least a little taken aback by that.

Point taken. Still, in the original SC the revelation that she turned gooey was the greatest thing ever. The eventual healing - well, not so much because it's to be expected right from the moment Jimmy looks at that picture in the bar. Maybe I am just disappointed that Blizzard took the obvious route this time." @atomic_dumpling: I don't think so. Kerrigan being healed in addition to the protoss prophecies creates a lot of tension for the games to come, because it leaves you with the question of how they are going to stop the Xel' Naga if she is not in Zerg form, yet she is technically also not dead and still a pretty powerful psychic so you never know. Mass Effect Overlord ending indeed, and Lair of the Shadowbroker, those scenes with Liara were so great that it made me sad.
Wish I could tell her that Garrus was just a blowing off steam for the suicide mission thing, but sadly that option wasn't available :/

NIER spoilers below"The way Emil's story ends in NIER was pretty sad, I really didn't like him at first but all the crazy shit that he ends up doing made me sympathize with him. His final sentence was pretty depressing. There was also the Popla and Devola death scene, Oh, and the king of Faceade wedding and everthing with Kaine...Man NIER was messed up!
@Dancrane play all the endings and perhaps the truth will set you free of your sadness. There's more to the story than the first ending. :p
What the hell was so sad about the end of Red Dead? Jack avenges his father. Boo fucking hoo. The fake ending was much sadder.
Ethan was shot to ribbons in front of his son after the girl (sorry, its be so long since I played I can't remember names) failed to inform him of what lay outside, the detective fell to a gruesome death and Shelby got away with it all.
Words can't describe the crushing feeling I had going to sleep after that.
I agree with the others who said Deadly Premonition.
Also, the way Shinjiro's Social Link ended in Persona 3 Portable got me down, not because I couldn't see it all coming, but...
