Director: Christopher Nolan - He can do action and make it thought provoking.
Not sure about actors. York should be played by somebody who can act really goofy and weird, but also seem totally serious whilst doing it. And be able to grow a beard in real time. I can't really think of anyone specific though (perhaps a young and up and coming actor would be best).
Game: Max Payne
Director: Robert Rodriguez (with Tarantino directing the dream/drug sequences. Wait, scratch that, I want David Lynch to do those.)
Lead Actor: That dude looks the part and I've heard good things about Six Feet Under.

Then again, 40 Year Old Virgin is pretty much as close as I'd like to get to a LSL movie, so there you go - it practically already happened. Still, no one does lovable losers quite like Apatow, and I loved their work together, so I wouldn't mind seeing 'em team up again with one of my favorite games.

Also he is way better a director than Tim Burton has ever been. Don't get me wrong I love Beetlejuice and A Nightmare Before Christmas certainly took up a lot of my childhood, but he is all about his movies just being creepy, that's all he has. del Toro has a great style that he delivers without the use (or primary use anyway) of CG.

I'm not a giant fan either I just tried to think of a badass chick off the top of my head and she seemed to fit the part better than Milla Jovovich :P
