Mii Support is the idea that your Mii characters created in the Mii Channel will make appearances in the game itself.  After the Miis are made you can generally see them in the background of some games.  In other games you can even interact with the Miis or play as them. Some games like Wii Sport are all about using your Mii.

The example of Mario Kart Wii

Mario Kart has always been about the racer's weight class. Heavy racers have slow aceleration and good top speed, opposit for lightweights and middle weight racers are in between. With Mii racers in Mario Kart Wii, the big question was if the settings you had choosen for your Mii would come into play (weight, height). What the game did was ignor all this and just make a Mii a middle weight racer (like Mario). Every Mii would race the same way. This shows that in most cases, Miis don't change the gameplay and all play the same.
