I haven't played Puzzle Agent, but they seem to be similar. I can definitely recommend the Layton games though. I would say charming is probably the best way to describe the tone of the Professor Layton games; most of the puzzles are good, some can be a little weird though.
I thought I'd enjoy Layton because I really liked Puzzle Agent, but I couldn't really get into it. I think without the interesting characters and story, I can't really play a puzzle game.
