    If a  mouse isn't necessary, is it practical to play the game with arrow keys?

    You definately need a mouse.  For casting spells and aiming beams more than movement though.

    There are setting options to play with a controller, but I can't get it to work, myself.

    The controller settings are pretty awful when it comes to making spells so you aren't missing much.

    When it comes to stacking and casting spells with a 360 controller all the 8 magics are laid out around your right joystick in pairs. As an example, fire and ice are on your right and when you want to stack fire you need to press the joystick first RIGHT then UP and let go. That'll stack fire once. I actually didn't find it all that bad. After a while you'll stack 5 magics in a second or two.

    I didn't try keyboard and mouse but I found it comfortable to play with the gamepad.

    I really doubt it. Not to say that the gamepad may not be serviceable, just that m+kb will be better/easier.
