I would rather like a reboot.
The main story ended nicely in RE5, and one of RE biggest problems was coming to conclusions with it's fiction. It went on way too long. Wesker was the bad guy who's thread connected the entire series, his final battle with Chris was an excellent way to end the series, or at least the main plot-thread. His plans are destroyed, Umbrella is destroyed, and now Chris and company can finally rest easy.
Either retell the story with more coherency, or do something drastically different. The idea of making a RE6 in the same universe with Wesker coming back from the dead like a comic-book villain doesn't seem appealing to me at all. Yeah, he "died" and resurrected in the first game, but that was the synapse of his origin to becoming the villain we all now and love. He died in a fucking volcano in the fifth game (>Insert Tekken 2 joke here<).
Make it permanent.
There's still the mysterious Organization. So far they have collected every single virus made by umbrella and both the standard and control plagas, I doubt they're just gonna sit on their @sses with all these biological weapons.
