Despite the relentlessly long presentation, were you still left wanting?
Given the state of Marvel vs Capcom, and no hints at all of a God Hand re-release in the future, this may have been Gene's last chance at appearing in a video game.

@flameboy84: Since this is the Wii U game ported to Switch, but with the additions required to consider it the "Ultimate" version of Smash for past players, i really think this is it.
Obviously they designed and created a whole new character in the form of Ridley, but given the "update" nature of the game i don't think they'll have plans for more new characters, at most maybe they have another Echo character or two.

It's not a port. In interviews it's been described as a "New game built specifically for Nintendo Switch."

Inklings are new as well. We already knew about them from the reveal trailer, but they are new for this game. I'd expect maybe a total of 5 new characters, so that would mean 3 more to be revealed. There's no way they haven't saved some announcements for the next 5 months between now and when it comes out.
I also think a subspace emissary style single player mode is likely. That Ridley reveal scene had way too much production value to only be a character trailer. My bet is that's a cutscene from the single player.
I'm pretty damn happy with the roster, plus Sakurai did say not to expect 'to many new challenges' so I'm guessing we'll get one or two more new challengers... maybe even three. Plus I think they'll add downloadable content to this game as well considering how well the Wii U/3DS DLC did.
Personally I'm a little disappointed Waluigi still isn't on the main roster. But I could totally see some form of Monster Hunter coming to Switch given the recent history between those two franchises.
ISAAC! At least let him be an assist again!
YO. I was legit disappointed they didn't show him when going through assists, I just want him to be acknowledged!

To answer the question in your OP... no one is missing; the game features every character ever released in a Smash Bros game.
I am going to assume what you meant to ask which character we would like to see. I'd like to see an iconic character from a Squaresoft game... Frog, Crono, and Geno in no particular order. That is my pick.
Pour one out for your boy, Bomberman. Christ look how good he looks as an assist trophy. I died a little on the inside when I saw that.

Like, sure there are other people who might make a Smash game one day. There are a very stand out absences that might be fun to see.
But taking the question at all seriously (and not the goof it should really be seen as) How is WaLuigi not just a..whatever it is, alter? if Luigi? They could have made it a purely cosmetic change and there you go, done. Most/if not all the other characters who might show up on a list of "Who's missing" would require a lot more work, whole move sets and etc. WaLuigi would require a single remodel of a character. Not no work but come on..
It really feels like someone at Nintendo is actively choosing to give his fans the middle finger. Like, they arn't, again this isn't -really- a serious topic. But its hard not to scratch your head when you consider the ease of adding him compared to other new characters, the good will it would bring, and how instead they did choose to show him off "Look he's just an assist trophy and you can kick his butt for a point haha!"
Simon Belmont feels like the last legacy NES character from a non Nintendo published game that's a glaring omission right now. Castlevania had its start on the NES and Belmont could easily fit in the roster. Goemon can too since his Finally Smash can be Impact, or at least give us an excuse for us to listen to his song again!
If Nintendo can get a deal with Microsoft, Banjo feels like someone who should be on that roster too. Smash is lacking of significant N64 reps and Banjo was a staple to the system.
