Rather than say movies everyone already knows, I'm gonna go with (in no particular order) ones that aren't as well known (Assuming you know the Coen Brothers) by kids these days, so as to educate your punk asses.

6. What's Up Doc is pretty good from what I remember, but that was like 8 years ago so don't hold me to it.

Zach and Miri make a Porno, Superbad, Forgetting Sarah Marshall, The 40-Year-Old Virgin, Pineapple Express, and Bruno are some of my favorites.

All three of the Monty Python films, Planes, Trains, and Automobiles, the collective works of Mel Brooks, and Airplane just to name a few.
With all of the answers I've seen so far you guys need to go back and watch some older films.

I must confess that a lot of the choices you guys have come up with have really disappointed me. To each his own I guess, but seriously, You Don't Mess With The Zohan? Aww c'mon! And to list it next to the masterpiece that is Dumb and Dumber? My heart weeps softly.
- A Day at the Races (1937) - Marx Brothers classic full of great one liners from Groucho.
- The Thin Man (1934) - William Powell and Myrna Low exchange jokes while trying to solve a murder case.
- One Two Three (1961) - James Cagney deals with the Cola-cola heiress's matrimonial adventure in a divided Berlin.
- His Girl Friday (1940) - Cary Grant & Rosalind Russell trade quips in this dialog heavy story of journalism.
- Arsenic and Old Lace (1944) - Cary Grant deals with his aunts' nefarious hobbies
- Some Like it Hot (1959) - Jack Lemmon and Tony Curtis hide from mobsters after witnessing a crime
- Annie Hall (1977) - Woody Allen dates Diane Keaton in New York City in the late 70's.
- The Blues Brothers (1980) - John Belushi & Dan Aykroyd try to get their band back together to perform a benefit for an orphanage .
- Talladega Nights: The Ballad of Ricky Bobby (2006) - Will Farrell races against Sacha Baron Cohen in NASCAR.
- Blazing Saddles (1974) - Cleavon Little is the first black sheriff in a small Western town
These may not have the strongest laughs throughout, but they never loose their charm:
- Wayne's World
- Young Frankenstein, Space Balls, & Robin Hood: Men in Tights (Mel Brook's finest works)
- Heavyweights
I refuse to answer since what people consider funny differs ALOT from each other and its just not worth it to even discuss it.
some of the things some people find funny, make me simply hate them, but then again i could be the one not getting it right..
its just pointless.
Newer style comedy sucks so much. Bring back the Mel Brooks and the Montey Pythons!
My top 5 would be
1. Silent Movie
2. Life of Brian
3. Pure Luck
4. High Anxiety
5. Cannonball Run 2
Not necessarily the best comedy of all time, but the funniest comedy of all time? Airplane! This is the funniest movie I have ever seen. I also like Dumb and Dumber, The Naked Gun (all 3 of them), South Park: Bigger, Longer & Uncut, Jay and Silent Bob Strike Back, Wedding Crashers, Ace Ventura: When Nature Calls. ;)

In no particular order, the movies that always make me laugh are Happy Gilmore, Billy Madison, Home Alone 1 &2, Jay and Silent Bob Strike Back, Dumb And Dumber, Major Payne and Kung Fu Hustle. Each of those has at least one moment that will make me laugh when I think about them
Spinal Tap. Probably the funniest and most quotable movie ever made.

I was going to go with just one and this is it:
It is the Citizen Kane of comedy. The political satire involved is sublime and utterly timeless. Everyone should see this film. The Marx Brothers are probably the funniest film comedians ever, and this is a peak for them.
The Marx Brothers made my list, too but I went with A Day at the Races because it contains my favorite scene from any of their movies. It just cracks me up every time I see it. YouTube LINK. The scene at the track's betting windows where Chico convinces Groucho to buy all these books to help him pick a winner. I just laugh at the perfect timing involved with each joke.
