I have been a massive sci-fi fan ever since I was a kid.  Back then, sci-fi was truly amazing as cgi didn't exist and science was still simple.  So starting out with Lost in Space, I moved into Star Trek, Space 1999, Battlestar Galactica and so on.

So, I want a discourse on the best sci-fi show ever made, but I want to restrict it.  Some shows just are not sci-fi in the traditional sense of the meaning.  Buffy and shows like it are fantasy, not sci-fi.  3rd Rock is comedy/sit-com, not sci-fi.  Cartoons are just cartoons.  Superheroes are just superheroes (so no to Heroes, brilliant show that it is).

Sci-fi has to be set in some futuristic setting or have alien type connotations.

Having watched so many sci-fi shows, I actually don't have any problems picking the best one.  The best one is:

What truly set Babylon 5 up as the best was that it was fully written before filming started.  It had 5 seasons that had a beginning, a middle and an end.  It avoided the problems that beset shows like Star Trek and Stargate (which I do enjoy by the way) where they just write more episodes after they get approval.  They have no direction, no point and eventually get boring.

The best thing about any Star Trek, but especially the original and Next Generation, is to watch the extras on the deck doing stuff to make them look busy.  It is really bad and terribly funny.

Original Star Trek. Maybe it is just that when I was a kid there weren't many of these shows. There were a couple of British imports like Space 1999 and UFO, but Star Trek, which was in reruns, I am not that old, was just so different and cool. My friends and I all bought the paperback books with the shows made into short stories in them. The cover are ruled.

I'm not sure what's funnier.  The fact that the fight scene was complete and utter shit or the fact that the lizard man (apparently Gorn) repeated "uuhhhhhLAHAY!" over 9,000 times.

I still haven't seen all of Firefly and I never got past the mini-series for the newer Battlestar (saw all the original ones though including Galactica 1980)

I'm surprised no one has mentioned Space: Above and Beyond yet.  That series suffered premature cancellation from the Fox Network.  Like Firefly it was cancelled way before it had a chance to find the right audience.

All the Star Treks but the first, Stargate, and a few other I can't recall. Don't have a favourite as I love them all equally. My guilty pleasure is Andromeda. I know it's bad, but I love it.

Let's see, I have been watching oh-so much of Stargate Atlantis and Universe lately, but I still do not think that tops Next Generation for me. There was something about the moral choices, interesting planets, and great story that kept me into that show. So, I guess I'll just make a list.

The correct answer is lost. No prime time show has been able to fully captivate its viewers for 6 years when all it started as a show of survivors on an island of mystery. Through those six years it became a science fiction show through the evolution of its story telling and the places they went and the things characters did.

Farscape is a kids' show and vomit inducing to anyone introduced to it over 13.  Babylon 5 got good when it introduced the political arch.  The first series was dreadful.  Loved Sliders.

Too many good ones to be able to limit yourself to having to pick just one. Sadly I think it's going to be a while before something new pops up though as there seems to be nothing on the horizon to fill in the sci-fi tv series beyond a few possible new shows coming to the BBC America later this year.
