Hound Tall is one of Moshe Kasher's podcasts, where he has two comedians on to discuss a topic with an expert in that field. They've had on Reza Aslan to talk about Islam in America and have talked about death and polyamory in the past.
Someone else has mentioned Harmontown, which is also great. It can get a little preachy, but I can't think of many other podcasts that are as naturally and easily funny.
