I know one that's pretty funny to listen to, it's called My Brother, My Brother, and Me. They give out advice for anything. Anything. I know Buzz_Clik is an avid listener. In fact, I found out about it through him.

I'm with plasma hear, there are some great podcasts from Kevin Smith & friends. Smodcast is, in my opinion, still the most funny (Kevin Smith and producer Scott Mosier talking shit about everything and nothing). But shows like Tell em' Steve Dave, Jay and Silent Bob Get Old, and Highlands: A Peephole History are also great. They basically have a podcast for every day of the week, so I'm sure you'll find something you like." The entire SModcast series on smodcast.com are fantastic. "
You could also check out the Audiobooks from Ricky Gervais, though those aren't free through itunes (though you can always procure them in some other way..).

Although it is no longer running, GFW Radio. Probably my all time favourite podcast. The episodes titled LAN party and PC project aren't really good because Jeff Green and Shawn Elliot had left the podcast by then. Anyway you can find them at http://www.1up.com/do/minisite?cId=3148397

Uhh Yeah Dude is the best podcast imo. Not everyone will like the humor but if you can get into it it's amazingly funny.
Also if your craving video game podcasts check out the old eps of Idle Thumbs its the only game podcast I've heard that comes anywhere near the greatness of the bombcast.
For gaming I like The Bombcast, Weekend Confirmed, The CAGcast (CheapAssGamer podcast), Bitmob and one most people don't know too well, Orange Lounge Radio. It's been on the air for 6 years week after week. It's like a lot of the others in format, but I like the crew. It runs about 2-1/2 hours each show. For non-games, I suggest tech podcast, Buzz Out Loud from CNet. And for sports and entertainment, I've been a fan of The Tony Kornheiser Show for years now. Sports is the foundation but it covers all other topics as well like movies, politics and whatever the host finds interesting. It's often quite funny.

I giggle like a little girl every time I see a new Rogan is up. One of the more interesting dudes out there. I like Carolla mostly for the rage. His rants are classic.

I've never laughed harder at anything out there, and will often rewind/replay entire podcasts because of the humor. Those are the ones I listen to at least semi-regularly (weekly listens: Bombcast, Gamespy Debriefings, Engadget, MBMBAM, Rebel FM. Listen to most of them while I work out. Can usually get through one each night.).
