I think that idea would play out very poorly. The Tomb Raider games may have been a bit over-the-top in a video gamey kind of way, but to nowhere near the extreme Duke Nukem was, nor were they as heavily based around the novelty of the main character. If Duke is to be reinvented, there are two problems to deal with; firstly, the kind of Schwarzenegger-esque action movie hero and ridiculous sci-fi the Duke Nukem games embody doesn't really exist in the current age, and secondly, Duke needs to look more pathetic as a character.
With Forever it was hard to tell to what degree 3D Realms and Gearbox understood that they were making their lead character look very dislikeable, but when you have your protagonist making bad jokes, acting like an arrogant dick, and even cracking wise about the rape and brutal death of people he knows, and that game makes no acknowledgement what he's doing is distasteful or even pats him on the back for it, that's not entertaining, that makes it look like your lead character is a complete sociopath and that you don't understand that what's going on is just uncomfortable.
If Duke is coming back the world around him needs to treat him as the sad human being he is, and there may also be some mileage in reinventing him to be a caricature of macho video game heroes, as opposed to a parody of dated action movie heroes. Perhaps there's room for a somewhat serious component in Duke Nukem, if that component is the message that a lot of the super-macho attitude that many games assume is often pretty immature and is not always the most appealing thing.
