Uncharted. As it is probably the franchise I enjoyed more, with the complete rundown of the relationships between the characters, gorgeous locations and setpieces.
And which is it for you?

Several people before you did in fact mention Mass Effect.
You can add another, as I will say Mass Effect as well. I would also say Bioshock, but Bioshock 2 can go die a horrible, painful death. Infinite looks pretty awesome though.

Sorry didn't notice that, but I only see two people who have.
The problem I have is there are so many! Though bioshock, uncharted, infamous and mass effect stand out to me
Dead Space - it's incredibly original and near flawless in execution of a genre that is plagued with poor controls and awful gameplay.
I love the Portal games. The endgame of Portal 2 can be a bit frustrating--I thought the goo worked against the spirit of that franchise a bit--but the original Portal might be the closest thing to a perfect game I've ever played.
Mass Effect is an obvious selection, in spite of the ire Mass Effect 3 has stirred. Someone mentioned the Arkham games, which is a smart choice.
It might very well be Portal, when I stop to think about it.
I'm going to be a little different and say Rock Band. The main titles always try to add something new with each game (better options of sorting your music, pro-modes, keyboard), it's fun to play on your own and there's always new music coming out either through weekly DLC or the Rock Band Network so you never run out of new music to play on each instrument, and it's an absolute blast to bring out at parties.
I feel that Bioshock, Gears of War, Portal, and Dead Space are the best IP's that got their start during this generation. I haven't played Assassin's Creed, Mass Effect has too many gameplay issues, and pretty much all of those PS3 exclusives are "meh" to me. Yes, I can't seem to make myself play Uncharted, InFamous, or the first two Resistance games. Resistance 3 is the exception rather than the rule.

Also Darksiders, I've only played one Zelda game and it wasn't to completion so everything Darksiders did was sort of new to me.
