I would say it's between L4D2 and MW2. But I'm leaning more towards MW2.

Street Fighter 4 is probably the one game this year that will truly last the test of time; it's a masterpiece.
*perhaps Dragon Age too, but I'm not really into that type of game.
If AC2 would have had just a tad bit more...flare I guess, that would be my pick. I LOVE that game. I'll go as far as saying I love it more than my pick, Uncharted 2. I know that sounds weird, but I just enjoyed playing AC2 more, but I think Uncharted 2 is a better built game. I don't know if that makes sense, but that's that.
Uncharted 2. It's the only game on a list of great games this year that makes me unusually excited for the future of what videogames can be visually, through storytelling, and solid gameplay design and pacing. Other games are great at one or two of those things, but Uncharted 2 nails them all save for a few small flaws.
Out of the games I have played, the one that stands out in my mind right now is Assassin's Creed II.
I dunno man, its hard. Currently i'm playing Assassin's Creed 2, and I dont know how many hours ive pumped into it...10? 15? I keep getting sidetracked with getting treasures, codex pages, all sorts of side quests... basically I get to a town or city, whatever, get all the viewpoints, buy the treasure map, and then its a million different icons of stuff to do. Story wise im early into venice, and thats all Ill say, so I dont know how long till the end, but man, awesome game.
I absolutely loved Borderlands (need to get the DLC once im done with assassins), got a level 50 character finishing it twice, and a couple of level 25s too. Another game where I pumped way too many hours into. Dragon Age was amazing, that was another 50 hours well worth my time... I actually really feel like playing again with a different class, which I rarely do once i finish a game.
and of course Uncharted 2, which was freaking amazing, amazing graphics, AMAZING voice acting (i liked the voice acting more than the graphics >_>), cool story, GREAT set pieces... just one of the best single player rides in a long time. The only downside for me, it was a bit too short for my taste (for all the hype it had about being so much longer than the first, I finished it in about the same time, 9 hours or so)
Batman, MW2 (single, i dont play multi), Infamous, Street Fighter 4, Ratchet Crack In Time, Shadow Complex were also all games I loved... I hate choosing just 1. In fact thats my top10 2009 games right there. GOTY? I dont know...im leaning towards Assassins tbh, we'll see after I finish it. its one of the first 4 I mentioned for me (AC2, Borderlands, DA:O or U2)
Because I buy/play games 6 + months after they've been released, I'm always behind games-wise (for example, I'm just finishing up Mirror's Edge, and I recently closed the books on Professor Layton & the Curious Village). So that means I haven't played any of the big hitters yet, like MW2 (which I'm 99% sure I won't like), Uncharted 2 and others.
Killzone 2 is amazing, but I haven't finished it yet. So for right now, I'll say Street Fighter IV because it got me ridiculously hooked on fighting games again.
Can I pick Persona 4? I played it this year, so I'm going with that if I can. Otherwise I would have to pick MW2.
I would have told you going in to the year that it would be MW2, but it simply hasn't grabbed me the way it's predecessor did, so going off of the absurdly high enjoyment rating and the time played factor, with a soupcon of out-of-left-field flavoring...
