Yes, several...sadly. I've written about each extensively over at my blog, but I'll summarize them here as well.
First, in Saints Row: The Third, couldn't finish the Deckers virtual reality stage that Giant Bomb praised so much because the dude Matt Miller would just fly up after taking X amount of damage and leave the screen entirely, leaving you stuck to run around an empty room, with no boss to kill (http://grindingdown.wordpress.com/2012/01/07/that-httpdeckers-die-glitch-where-the-final-boss-simply-disappears/).
Second, in Half-Life 2, there's a switch you are to flip in the Nova Prospekt jail level to open a door (http://grindingdown.wordpress.com/2011/12/05/switch-glitch-in-nova-prospekt-level-brings-half-life-2-to-a-halt/). Problem is that there is no switch for me to push. Well, not true. The switch is there, it just won't activate no matter how many times I try. Ugh. I've read online that I might need to restart the game, but I don't wanna do that.
Third, in Deus Ex: Human Revolution, right near the end of the game, there was a set of double doors that, when looked at directly, would freeze the game (http://grindingdown.wordpress.com/2011/10/19/ridiculous-door-glitch-in-deus-ex-human-revolution-is-ridiculous/). I kid thee not. If you were able to look at the floor and still open the door, you could pass by safely. But raise your eyes an inch, and everything locked up. That took a bit of trial and error to get around, if you couldn't already tell.
And let's just consider most of Fallout 3 and Fallout: New Vegas is one big progression-blocking bug, unless you save every single step of the way.
